To provide a novel tricyclic thiophene compound that specifically enhances the action of a cell differentiation-inducing factor in vivo and is useful as a prophylactic or therapeutic agent for osteopathy and neuropathy, particularly for prophylactic or therapeutic agent for osteoporosis, or as an osteoplasty promoter in the restoration and transplantation of bones or alveolar bones.
This novel compound is represented by formula I [R1 is a lower alkyl; R is cyano, carbamoyl; A is a (substituted) aromatic five-membered ring; n is 3-10], typically 9-methylthio-4,5-dihydro-6H-1-oxa-8-thia-2- azacyclopenta[e]azulene-7-carboxamide. The compound of formula I is prepared, for example, by brominating a compound of formula II to a compound of formula III with a brominating agent, then cyclizing the compound of formula III with a thiocarboxamide or a Schiff base to a compound of formula IV or formula V.
