Abstract:
Tumor-associated peptides (I) having any of 79 sequences, reproduced, are new. (I) can bind to a Class I molecule of the human major histocompatibility complex (MHC). Independent claims are also included for the following: (1) nucleic acid (II) that encodes (I); (2) vector that contains (II); and (3) cell modified to express (I), by incorporation of (II) or the vector of (2). ACTIVITY : Cytostatic. MECHANISM OF ACTION : Vaccine; induction of cytotoxic T lymphocytes (CTL). Dendritic cells (0.5 million) were isolated from peripheral blood and incubated with peptide (1) at 50 mu g/ml for 2 hours, then washed and incubated with 2.5 million autologous peripheral blood mononuclear cells (PBMC) for 7 days. The cells were restimulated with interleukin-2 (1 ng/ml) on days 1, 3 and 5, then cytotoxicity tested against various cancer cells at effector:target ratio 30:1, in a standard 51chromium release assay. Typical specific lysis scores were over 30% for HCT 116 stomach cancer cells; over 60% for A498 renal cancer cells; about 40% for MEL1479 melanoma and about 50% for MCF-7 breast cancer cells. Tyr-Val-Asp-Pro-Val-Ile-Thr-Ser-Ile (1).
