PURPOSE: To hybridize a nucleic acid probe after amplification of a nucleic acid in a sample containing a specific exon of HLA Class I allelic gene and to type HLA Class I-DNA for diagnosis, forensic medicine, etc., from the resultant pattern.
CONSTITUTION: In a method for typing HLA Class I-DNA of a nucleic acid in a sample, the nucleic acid in the sample containing Class I-HLA allelic gene second and/or third exon sequence is respectively amplified, the second exon sequence is amplified with a pair of primers composed of formulae I and II, and the third exon sequence is amplified with a pair of primers composed of formulae III and IV, and the amplified nucleic acid is hybridized with an oligonucleotide probe of a first panel of formula V, etc., under a condition that the probe hybridizes to only correctly complement sequence, then a Class I-HLA allelic gene in the sample useful in a field of medical research and diagnosis, a field of forensic medicine, and in a field of molecular biochemistry is decided from the resultant pattern of probe hybridization.
