Abstract:

To provide methods of attenuating, for example, inhibiting or reducing, cellular proliferation and cellular migration, particularly endothelial cell proliferation and endothelial cell migration that are associated with angiogenesis.

In a treatment method, opioid antagonists, such as naltrexone and alvimopan, are used which may be, but not limited to, peripherally restricted antagonists, wherein the treatment method including administering the opioid antagonist solely or by the combination of the antagonist and anticancer drugs, such as 5-fluorouracil and bevacizumab, is effective for such disorders as a cancer, diabetes, sickle cell anemia, blood vessel damage, and proliferative retinopathy. The effective amount is such that a subject has effective circulating blood plasma levels of the opioid antagonist continuously for at least 1 week, at least 2 weeks, at least three weeks and, preferably, at least 4 weeks.
