Abstract:

PURPOSE: To enable an increase of half-life in blood and an improvement in thrombolytic ability by substituting a specific amino acid residue mainly in an EGF region of a human PUK which is a fibrinolytic enzyme.
CONSTITUTION: The objective variant human prourokinase is obtained by substituting Ser, Asn, Pro, Gly or Tyr in a region from Asn at the 10th residue to Gly at the 60th residue on the N-terminal side of a human prourokinase (human PUK) with Thr, Pro or Ala. Furthermore, a method for producing the human prourokinase characterized by culturing a host transformed with the variant human prourokinase and expressing the variant human prourokinase is provided.
