Abstract:
PURPOSE: To obtain a variant type streptavidin gene, capable of coding an amino acid sequence of a specific variant type streptavidin and efficiently producing a fusion protein of streptavidin-firefly luciferase and useful for the immunoassay, detection of a DNA, etc.
CONSTITUTION: This variant type streptavidin gene has an amino acid sequence expressed by formula I at the N-terminal and is capable of coding an amino acid sequence expressed by formula II. Furthermore, this fusion protein gene of the variant type streptavidin-firefly luciferase is obtained by joining the gene to a firefly luciferase gene. This method for producing the fusion protein of the variant type streptavidin-firefly luciferase is to culture a microorganism, belonging to the genus Escherichia and containing a recombinant DNA prepared by inserting the fusion protein gene of the variant type streptavidin-firefly luciferase into a vector DNA in a culture medium and collecting the fusion protein of the variant type streptavidin-firefly luciferase from the resultant culture.
