Abstract:

PURPOSE: To obtain the subject compound useful as an intermediate for compounds having the antiulcer activity in high purity and yield without using an expensive raw material.
CONSTITUTION: 4-Chloro-2-chloromethyl-3,5-dimethylpyridine-N-oxide expressed by formula I is made to react with 2-mercapto-5-methoxyimidazo[4,5-b]pyridine expressed by formula II in a solvent, preferably ethanol at a temperature within the range of 10°C to the boiling point of the solvent, preferably at 30 to 50°C temperature to provide 4-chloro-2-[(5-methoxy-2-imidazo[4,5-b]pyridinyl)thio]methyl-3,5-dimethylpyridine-N-oxide expressed by formula III. The resultant compound is then allowed to react with sodium methoxide in a molar amount of 2.0-10.0 times that of the compound expressed by formula III in the presence of an organic solvent to afford a compound expressed by formula IV, which is subsequently made to react with phosphorus trichloride or pentachloride in a molar amount of 2.0-10.0 times that of the compound expressed by formula IV in the presence of an organic solvent to provide the objective compound expressed by formula V. The obtained compound expressed by formula V is then oxidized to afford a compound, expressed by formula VI and having the antiulcer activity.
