Abstract:
Pancreatin pellets (1), preferably micropellets, comprise a 100% pancreatin content obtained from the pancreas of a mammal and having a spherical, ellipsoidal or drop shape and an axis of 0.5-2.5 mm. The pellets are produced by (a) comminuting pancreas glands of pigs or cattle and carrying out an autolysis, (b) obtaining a screen filtrate containing enzymes, (c) precipitating the enzymes, (d) filtering the mixture, (e) cooling the filter cake, (f) extruding the cooled filter cake, (g) spheronizing and (h) vacuum drying the pellets. Pancreatin pellets (1), preferably micropellets, comprise a 100% pancreatin content obtained from the pancreas of a mammal and having a spherical, ellipsoidal or drop shape and an axis of 0.5-2.5 mm. The pellets are produced by (a) comminuting pancreas glands of pigs or cattles and carrying out an autolysis, (b) obtaining a screen filtrate containing enzymes by filtering the intermediate product obtained after the step (a), (c) precipitating the enzymes out of the screen filtrate, (d) filtering the mixture obtained after the step (c) to obtain a filter cake, (e) cooling the filter cake obtained after the step (d) to -10[deg] C to -40[deg] C, preferably to -30[deg] C, to obtain a sufficient plasticity, (f) extruding the cooled filter cake obtained in the step (e), and excluding additives and/or binding agents to form extrudate, where the extrudable filter cake mass contains residual moisture or organic solvent residues up to the order of 30-50%, preferably 40%, (g) spheronizing, and excluding additives and/or binding agents or other auxiliary materials to obtain spherical, elliptical or drop-shaped pellets and (h) vacuum drying the pellets obtained until the residual moisture content is less than 3 wt.%, preferably less than 1 wt.% or less than 0.5 wt.% to obtain a dried pancreatin end product with a residual moisture content of less than 3 wt.%, preferably less than 1 wt.% or less than 0.5 wt.%. Independent claims are also included for: (1) preparing pancreatin pellets, preferably micropellets by the above mentioned method; and (2) a pharmaceutical composition comprising the pancreatin pellets.
