Abstract:
Provided is an α-amylase that exhibits excellent starch degradation activity at a low temperature and excellent stability at a low pH and/or in the presence of a chelating agent. This α-amylase variant is any one of α-amylase variants A1, B1, C1, D1, and E1, which are respectively obtained from parent amylases that are: an α-amylase A having at least 90% sequence identity with respect to the amino acid sequence indicated by SEQ ID NO. 4; an α-amylase B having at least 90% sequence identity with respect to the amino acid sequence indicated by SEQ ID NO. 6; an α-amylase C having at least 90% sequence identity with respect to the amino acid sequence indicated by SEQ ID NO. 8; an α-amylase D having at least 90% sequence identity with respect to the amino acid sequence indicated by SEQ ID NO. 10; and an α-amylase E having at least 90% sequence identity with respect to the amino acid sequence indicated by SEQ ID NO. 12. The α-amylase variants A1-E1 include deletions of amino acid residues at two sites selected from positions corresponding to positions 178-181 in the numbering in SEQ ID NO. 1, and have substitutions that are in amino acid residues at three or more sites selected from positions 178, 181, 238, 239, 240, 126, 129, and 201 in the numbering in SEQ ID NO. 2 and that include at least a) to e). a) For A1, 178H, 181E or 181Q, and 239Q. b) For B1, 178H, 181E or 181Q, and 239Q. c) For C1, 178H, 181E or 181Q, 238F, and 239Q. d) For D1, three or more sites selected from 178H, 181E or 181Q, 238F, and 239Q. e) For E1, three or more sites selected from 178H, 181E or 181Q, 238F, 239Q, and 240F.
