Abstract:
Provided is an ancestral sequence reconstruction-based ω-transaminase mutant from Aspergillus terreus. The amino acid sequence of the mutant is as shown in SEQ ID NO: 4 or SEQ ID NO: 6. Compared with a wild-type enzyme, the thermal stability of the ω-transaminase mutant is improved. Specifically, the half-life of the mutant is 24 h or above, and the half-life of the wild-type enzyme is 6.90 min. The semi-inactivation temperature of the mutant is about 49.00°C, and the semi-inactivation temperature of the wild-type enzyme is 37.89°C.
