Abstract:
Disclosed are an anti-pancreatic cancer polypeptide drug conjugate, and a carrier-free self-assembly nano drug thereof, and a preparation method therefor and the use thereof. The anti-pancreatic cancer polypeptide drug conjugate takes an active oxygen response fragment as a connecting arm, and chemically conjugates the pro-apoptotic peptide SmacN7 and gemcitabine. The carrier-free nano drug is formed by means of self-assembling the polypeptide drug conjugate and a photosensitizer molecule chlorin e6 (Ce6) by means of a one-step nano-precipitation method. The provided carrier-free nano drug has the advantages of small and uniform particle size, good stability, etc., can release the chemotherapeutic drug gemcitabine and the pro-apoptotic peptide SmacN7 as needed under the illumination stimulation of 630-670 nm in tumor cells, synergizes the effects of photodynamic therapy and chemotherapy, can significantly improve an anti-tumor effect thereof, and has an excellent clinical conversion prospect.
