Disclosed are an anti-phenacetin monoclonal antibody hybridoma cell strain AD, and a preparation method therefor and the use thereof, which relate to the technical field of food safety and immunological detection. The deposit name of the monoclonal antibody hybridoma cell strain is monoclonal cell strain AD, and the deposit number is CGMCC19681. The hybridoma cell strain is obtained by means of hydrolyzing a reaction product of phenacetin metabolite paracetamol and ethyl 4-bromobutyrate to obtain Phe-BA as a hapten, coupling the hapten to a carrier protein to prepare an immunogen Phe-BA-BSA, injecting the immunogen Phe-BA-BSA to mice for immunization, then fusing with myeloma cells by means of a PEG method, and carrying out screening and carrying out subcloning five times by means of an indirect competitive enzyme-linked immunosorbent assay. A monoclonal antibody secreted by the cell strain can be prepared into a phenacetin detection kit, which has a good affinity for phenacetin and a good detection sensitivity thereto, and can be used for the immunological detection of the residual amount of phenacetin in food.
