Abstract:
Disclosed is an apparatus for measuring fluorescence signals in a plurality of illumination modes. Excitation light is modulated in an illumination excitation light path; excitation illumination patterns with different phases are generated at a sample (32) after the excitation light passes through an objective lens (31); a high-speed switching apparatus is provided in a fluorescence collection light path to switch the position, on a target surface of a photoelectric sensor (40), of a fluorescent image of the sample (32); and by means of the simultaneous operation of a plurality of illumination excitation light paths and a plurality of fluorescence imaging light paths, a plurality of sub-images, i.e. fluorescence signals corresponding to a plurality of illumination modes, can be simultaneously obtained after a single instance of exposure. Fluorescence signal measurement of a single molecule can be realized in the plurality of illumination modes, the precision of positioning a single molecule can be improved, and a higher resolution capability can be realized under the condition of having an equal number of photons. Further disclosed are a measurement method for the apparatus for measuring fluorescence signals in a plurality of illumination modes, and an application thereof.
