Abstract:
Provided is a transgenic artificial antigen presenting cell characterized by: (1) expressing a T cell co-stimulatory signal membrane-bound activating antibody, a target antigen and a cytokine; and/or (2) stably integrating an expression cassette containing codes of T cell co-stimulatory signal membrane-bound activating antibody, the target antigen and the cytokine in the genome; preferably, both ends of the expression cassette comprise an inverted terminal repeat sequence of the transposon. The artificial antigen presenting cell can induce the proliferation and activation of CAR-T cells, generate central memory CAR-T cells, stimulate CAR-T cells to secrete IFN-γ, and enhance the ability of the specific CAR-T cells to kill tumours.
