Abstract:
The present invention relates to use of SC79 or an analogue thereof in the preparation of a blood brain barrier permeability modifier. The blood brain barrier permeability modifier uses SC79 or an analogue thereof as an active ingredient to down-regulate the expression of closely linked proteins Claudin-5 and Occludin by Claudin-5 and Occludin activating signal pathways downstream of protein kinase B, thereby increasing blood brain barrier permeability, enhancing the transport efficiency of a brain-targeted drug delivery system into the brain, and particularly enhancing the transport efficiency of an Angiopep-2 modified glycolipid nano drug delivery system into the brain. The present invention also relates to a kit comprising the blood brain barrier permeability modifier and a brain-targeted drug delivery system. According to the present invention, by using said blood brain barrier permeability modifier and brain-targeted drug delivery system in combination, the transport efficiency of the brain-targeted drug delivery system into the brain can be enhanced, and the therapeutic effect of the brain-targeted drug delivery system can be improved.
