Abstract:
The present invention relates to a chimeric antigen receptor-modified T cell targeting Muc1 and the use thereof. In particular, the chimeric antigen receptor provided by the present invention, from the N-terminus to the C-terminus, successively comprises a membrane protein signal peptide, a single-chain antibody combating the membrane proximal end of Muc1, a hinge region of more than 50 amino acid residues, a transmembrane region, a co-stimulatory signal molecule intracellular domain and an immunoreceptor tyrosine-based activation motif. The present invention also comprises a T cell expressing the chimeric antigen receptor. The T cell can recognize and bind to the membrane proximal end of the Muc1 antigen, and kill Muc1-positive tumour cells by efficiently activating proliferation and secreting cytokines. The T cell provided by the present invention can be used for the treatment of various Muc1-positive malignant tumours.
