Abstract:

A DNA fragment of about 3.8 kb which is prepared by cutting hybrid plasmid pST 141 of about 12.6 kb, contained in Streptomyces griseus 4-1 strain (FERM BP-1198) with a restriction enzyme BglII and again cutting the resulting BglII.BglII-cut DNA fragment of about 7.0 kb with a restriction enzyme SphI, said DNA fragment having restriction enzyme sites shown in the restriction enzyme map of Fig. 1. This DNA fragment contains a streptomycin-resistant gene and a portion capable of regulating manifestation of the gene near the gene. When incorporated in a proper actinomycetes plasmid vector, the fragment can form a hybrid plasmid surely functioning as a streptomycin-resistant selected marker.
