A purpose of the present invention is to provide a filler for ion exchange chromatography, by which a difference in the nucleotide sequence in a nucleic acid strand or difference of single nucleotide substitution can be sufficiently detected. Another purpose of the present invention is to provide a method for separating and detecting a nucleic acid strand, which uses the filler for ion exchange chromatography. The present invention is a filler for ion exchange chromatography, which comprises a strong cationic group and a weak cationic group on the surface of a fine base particle.
