Abstract:
A novel fluorescence indicator for analyzing an intermolecular interaction or intramolecular structural change with the use of fluorescence resonance energy transfer (FRET). There is provided a fluorescence indicator having such a structure that a donor fluorescent protein and an acceptor fluorescent protein are linked to both ends of a target sequence for analyte wherein the three-dimensional structure of the indicator is changed by analyte liking to the target sequence or action on the target sequence to thereby induce fluorescence resonance energy transfer (FRET), characterized in that the donor fluorescent protein and/or acceptor fluorescent protein is a circular permutation variant fluorescent protein obtained by interchanging between N-terminus side amino acid sequence and C-terminus side amino acid sequence of wild type fluorescent protein or its variant protein and is a fluorescent protein having substantially the same fluorescence peak wavelength as that of the fluorescent protein before being subjected to the circular permutation variation.
