Abstract:
A focusing method according to one embodiment of the present invention is performed by a device that acquires image data of a biological sample. The method includes: a step for acquiring first image data items of the biological sample at a plurality of positions along an optical axis of an objective lens included in the device, and position data items pertaining the position, along the optical axis, of the first image data items; a step for executing smoothing processing on the first image data items so as to generate respective second image data items; a step for acquiring contrast values of the respective second image data items; a step for identifying a first peak value and a second peak value among the contrast values on the basis of the position data items; a step for, between the positions of the objective lens on the optical axis at which the first peak value and the second peak value among the contrast values are obtained, identifying a position on the optical axis at which a bottom value of the contrast values is obtained; and a step for setting, as a focal position of the device, the position on the optical axis at which the bottom value of the contrast values is obtained.
