Abstract:
A preparation method for a functional nanomagnetic bead for enriching potato ester acyl hydrolase, comprising the following steps: mixing a carboxylated nanomagnetic bead with ethyl-dimethylamine-propylcarbodiimide; activating the nanomagnetic bead; and coupling the activated nanomagnetic bead with aleuria aurantia lectin, so as to obtain the functional nanomagnetic bead. The present invention also provides use of a functional nanomagnetic bead coupled with aleuria aurantia lectin for enriching the potato ester acyl hydrolase, comprising the following steps: extracting crude protein from potato starch waste water; performing impurity removal and elution to obtain an eluate; using the functional nanomagnetic bead to enrich the potato ester acyl hydrolase in the eluate; and performing elution and purification to obtain the potato ester acyl hydrolase. By using the functional nanomagnetic bead prepared by the present method to enrich or adsorb and immobilize the potato ester acyl hydrolase, the time for enriching the potato ester acyl hydrolase can be shortened, the purity and enzyme catalytic performance of the potato ester acyl hydrolase can be improved, and the biological activity thereof can be retained.
