Abstract:
Provided is a gene transcription framework, a vector system, and a genome sequence editing method and application. The gene transcription framework is based on an eukaryotic retrotransposition mechanism and may be mediated by DNA, RNA, or RNP pathways, and comprised are a transcript sequence comprising a target site upstream sequence, a sequence to be inserted, and a target site downstream sequence, one or more Sine (elements), a LINE (element), one or more ORF1p coding sequences, and/or an ORF2p coding sequence. In the gene editing method, insofar as foreign systems or substances are as much as possible not introduced and no double-stranded breaks are generated, transfer into the nucleus or cytoplasm occurs by means of the DNA, RNA or RNP pathways (RNA or RNP is mediated to be transferred from the cytoplasm to the cell nucleus by means of the ORF1p and/or the ORF2p), and a target fragment is inserted into a specified site in the genome or a specified fragment in the genome is deleted or replaced.
