Abstract:
The present invention belongs to the technical field of preparation methods for polypeptides, and particularly relates to high-purity corticotropin (human-derived sequence) or an analog thereof, and a large-scale preparation method therefor, which method comprises the main steps of: firstly, coupling amino acids in sequence from a C terminus to an N terminus by means of an Fmoc solid-phase synthesis method to acquire a crude peptide-resin of corticotropin (human-derived sequence) or an analog thereof with a protective group, wherein the reaction temperature of C-15 peptide synthesis is 40-60ºC; then cutting and settling same to obtain a crude product of the corticotropin (human-derived sequence) or the analog thereof; and then separating and purifying same by means of liquid chromatography to acquire a high-purity finished product. The chromatographic purity of a sample of the corticotropin (human-derived sequence) or the analog thereof prepared by the present invention reaches 99% or above, the stability is good, and the yield of a target peptide is ≥63%.
