Abstract:
Provided are a human immunodeficiency virus (HIV) pseudovirus particle and a method for preparation thereof. The pseudovirus particle comprises a conserved sequence fragment of Gag, Pol, and LTR regions on an HIV-1 gene as a target fragment and a bacteriophage MS2 protein. The method comprises: designing and selecting a target fragment and a bacteriophage MS2 gene sequence; directly synthesizing the two respectively; inserting the bacteriophage MS2 gene sequence into a pETDuet-1 carrier to obtain a pETDuet-MS2 carrier; inserting the target fragment into a pETDuet-MS2 vector to obtain a pETDuet-MS2-HIV recombinant expression vector. The phage MS2 gene sequence and the target fragment are respectively located between any enzymatic cleavage sites behind the tail ends of two T7 promoters of the pseudovirus particle.
