Abstract:
The present application provides an integrated organ chip and a preparation method therefor, and relates to the technical field of biomedicine and microflow. The integrated organ chip is configured for applying electrical stimulation and mechanical stimulation as required in a process of culturing a cell, a cell cluster or an organoid. The integrated organ chip comprises an electrode layer configured for providing electrical stimulation, a microporous layer located above the electrode layer and configured for culturing, a flow chamber layer configured for providing mechanical stimulation and interacting with an external connection, and a peripheral circuit. The electrode layer comprises a substrate and bare electrodes which are located on the substrate corresponding to micropores in the microporous layer. The microporous layer comprises a structure which is adhered to the electrode layer and has a plurality of mutually independent single-pore chambers. Each single pore is permeable with the bottom corresponding to one bare electrode or a multielectrode consisting of a plurality of electrodes in the electrode layer. The flow chamber layer is a structure with a plurality of single cavities which is adhered on top of the microporous layer and configured for carrying a culture medium to which a culture belongs. By means of the method, various regulation and control technologies can be combined to meet the culturing requirements for the organoid or the cell cluster and promote function maturity of the organoid or the cell cluster.
