Abstract:
The present invention belongs to the field of gene sequencing, and particularly relates to a library construction and sequencing method for a single-stranded nucleic acid cyclic library. In the method, a sequencing primer binding sequence is combined in a linker molecule or the sequencing primer binding sequence is separately contained in the linker molecule and an extension reaction primer, so that reaction steps for connecting a sequencing linker are reduced, the loss of a nucleic acid template is reduced, the linker connection efficiency is improved, the utilization rate of an original nucleic acid template is improved, and rapid library construction and sequencing of various nucleic acid samples are achieved.
