Abstract:
It has been found that adhesion molecule CAR whose localization in intercellular adhesion sites is known functions as an adhesion molecule for activated lymphocytes. Further, a novel ligand CARL to CAR expressed in lymphocytes has been identified, and it has been elucidated that this ligand is selectively expressed in Th1 cells. Still further, it has been found that the adhesion of activated lymphocytes to CAR molecules can be inhibited by an anti-CAR antibody. Consequently, there is provided a method of detecting Th1 cells with the use of CAR or an anti-CAR antibody, and provided a method of screening an inhibitor capable of suppressing adhesion of Th1 cells with the use of binding between CAR and CARL as an indicator. Moreover, there are provided a method of screening an inhibitor for binding between CAR and CARL; an antibody capable of inhibiting any binding between CAR and CARL; and a therapeutic composition comprising this antibody. These would be useful for the diagnosis of diseases, such as inflammations wherein infiltration of Th1 cells gets involved, and for supply of medicine for alleviating the pathological symptoms thereof.
