Abstract:
It is intended to provide a method which comprises specifying a structural factor determining the fluorescence wavelength in the three-dimensional structure of a GFP-like fluoroprotein originating in Copepod and modifying an amino acid residue participating in the structural factor so as to change the fluorescence wavelength while sustaining the recombination expression efficiency thereof; and a modified fluoroprotein that is obtained by applying this method. For example, in a GFP-like protein originating in Chiridius poppei, His52 in the α-helix type secondary structure: PFLLSHCMGYGFYHF(α1 47-61) including a fluorescent group GYG is substituted by an aromatic amino acid selected from among Phe, Tyr and Trp to cause a color shift of the fluorescence peak wavelength toward red; or substituted by Ala, Val, Ile, Leu, Gly, Cys, Met, Ser, Thr, or Asp, Asn, Glu or Gln to cause a color shift of the fluorescence peak wavelength toward blue.
