Abstract:
The purpose of the method according to the present invention is to specifically detect G-quadruplex formation even in the presence of potassium ion. According to the method of the present invention, the presence or absence of G-quadruplex formation by a target DNA is determined utilizing a phenomenon that thioflavin T emits strong fluorescence when reacted with G-quadruplex in the presence of potassium ion. The method of the present invention comprises: maintaining a first sample solution, which contains potassium ion and the target DNA, under such conditions as allowing G-quadruplex formation; adding thioflavin T thereto; measuring a first fluorescence intensity; maintaining a second sample solution containing the target DNA under such conditions as making G-quadruplex formation unstable; adding thioflavin T thereto; measuring a second fluorescence intensity; and, in the case where the difference between these intensities is 0 or greater, determining the target DNA as being capable of forming G-quadruplex.
