Abstract:
A method for editing a target polynucleotide, comprising the specific steps of: designing a pair of DNA oligonucleotide single strands for a target gene of interest, such that the pair of single strands are respectively complementary to the sense and antisense strands of the target gene; respectively generating a structure recognizable by a type of nuclease which recognizes a target sequence structure and which has a cleaving activity, and cleaving a target genomic DNA by means of the nuclease, so as to edit a genome. The structure recognized by the type of nuclease which recognizes a target sequence structure and which has a cleaving activity is a 3'-Flap structure, the 3' ends of the DNA oligonucleotide single strands are not complementary to the target gene; a recombinant structure recognizes nucleic acid endonuclease and includes a structural recognition domain, a DNA cleavage domain, a peptide linking said two domains, and a nuclear localization signal.
