Abstract:
The present invention addresses the problem of providing a method of highly-sensitive detection of adeno-associated virus (AAV)-neutralizing antibody, and providing a tool (e.g., DNA, cells) used in said method. Specifically, the present invention addresses the problem of providing a vector composition for AAV-neutralizing antibody detection, the composition containing vectors of (a), (b) and (c) below. (a) A vector comprising a DNA that includes a 5' transposon-specific inverted terminal repeat (ITR) sequence (hereinafter referred to as "5'ITR sequence"), a promoter, a transcription termination STOP sequence that can be deleted by a site-specific recombinant enzyme, one or more reporter genes, and a 3' transposon ITR sequence (hereinafter referred to as "3'ITR sequence"), which are connected in this order, the DNA being configured such that if the transcription termination STOP sequence is deleted, the one or more reporter genes are expressed by said promoter (hereinafter referred to as "reporter gene expressing cassette"); (b) a vector comprising a DNA that includes a 5'ITR sequence, a promoter, a selection marker gene, and a 3'ITR sequence, which are connected in this order, the DNA being configured such that said selection marker gene can be expressed by said promotor (hereinafter referred to as a "selection marker gene expressing cassette"); and (c) a vector comprising a DNA in which a promotor, and a transposase gene that recognizes the transposon ITR sequences included in vectors (a) and (b), are connected in this order (hereinafter referred to as "transposase gene-expressing cassette"), the DNA being configured such that said transposase gene can be expressed by said promoter.
