Abstract:
This method for extracting nucleic acids from embedded tissue extracts nucleic acids from embedded tissue by: bringing at least a portion of the surface of the embedded tissue, which has been embedded using an embedding agent that is insoluble in water and has a melting temperature in the range of 40-90°C, into contact with an aqueous nucleic acid extraction solution containing one or more substances selected from a group consisting of surfactants and alkaline substances in a nucleic acid extraction vessel at or above the melting temperature of the embedding agent to extract the nucleic acids in the embedded tissue into the aqueous nucleic acid extraction solution; separating the embedding agent layer, which has the melted embedding agent as the main component, and the aqueous layer, which contains the extracted nucleic acids, from each other; after separating said embedding agent layer and the aqueous layer from each other, solidifying the embedding agent layer while keeping the aqueous layer in liquid form with a cylindrical member, which has an external circumferential surface shape that corresponds to the inner surface shape of the nucleic acid extraction vessel, set inside the nucleic acid extraction vessel so that at least the lower end of the cylindrical member contacts the aqueous layer and at least the upper end of the cylindrical member is not in contact with the embedding agent layer; and after the embedding agent has been solidified, removing the cylindrical member from the nucleic acid extraction vessel together with the solidified embedding agent layer.
