Abstract:
Provided are a method for improving the expression of lipase by glycosylation modification, mutant enzymes and an application of the mutant enzymes, specifically comprising: performing N-glycosylation mutation on a propeptide sequence of rhizopus oryzaelipase, and respectively modifying SAS and/or NT amino acids into N-glycosylation loci NGT and/or NLT; compared with the extracelluar protein concentration of proROL which is not subjected to glycosylation, extracelluar protein concentrations of obtained mutant enzymes proROLA, proROLB and proROLAB are separately improved by 211 percent, 188 percent and 233 percent; and when a fermentation tank is adopted for culturing, enzyme activities separately reach 8210U·mL-1, 8457U·mL-1 and 9366U·mL-1, and the extracellular enzyme activity of unmutated proROL is almost zero.
