Abstract:
The present invention provides a method for detecting monoclonal antibodies in a sample, the method including (a) a step for trapping monoclonal antibodies in a sample and immobilizing the same in pores of a porous substance, (b) a step for bringing the porous substance in which the monoclonal antibodies have been immobilized and nanoparticles to which a protease has been immobilized into contact with each other, and subjecting the monoclonal antibodies to selective protease digestion, and (c) a step for detecting peptide fragments obtained through the selective protease digestion by means of liquid chromatography-mass spectrometry (LC-MS), wherein the method also includes (a') a step for carrying out a reduction reaction under acidic conditions, said step (a') being performed after step (a). The present invention makes it possible to anticipate further practical application of methods for detecting monoclonal antibodies using mass spectrometry.
