Abstract:
Provided is a method for preparing menstrual blood-derived mesenchymal stem cells, comprising: sieving a menstrual blood sample to obtain a filtrate; performing density gradient centrifugation on the filtrate to obtain a separation solution with four layers: a plasma layer, a buffy coat layer, a separation liquid layer, and a blood cell layer; collecting the buffy coat layer and the blood cell layer for mixing; and performing adherent culture to obtain menstrual blood-derived mesenchymal stem cells, wherein the method for collecting the blood cell layer is a filtration method comprising the following steps: mixing the blood cell layer with a cleaning solution, filtering through a 40-70 μm filtration apparatus, discarding a filtrate, rinsing an intercepted substance with a cleaning solution, and collecting the cleaning solution containing the intercepted substance. The method can improve the recovery rate of primary target cells on the basis of an original traditional method, and the proliferation ability of the cells in the later culture process is stronger than that with a traditional separation method.
