The present invention relates to a method for preparing high-content and high-purity omega-3 triglyceride comprising synthesizing under vacuum conditions by using omega-3 fatty acid ethyl ester, glycerol, and enzyme. The method for preparing of the present invention is a method for efficiently mass-producing third generation omega-3 triglyceride, which has improved the problem of low concentration of an active ingredient of first generation omega-3 and the problem of stability and bioavailability of second generation omega-3. Specifically, the method for preparing of the present invention is a mass-production synthesis method that minimizes processing processes by using processed enzyme differentiated from previous experiments to improve reactivity, and by maximizing reaction efficiency under conditions differentiated from previous experiments, and may provide omega-3 triglyceride in which a purity of triglyceride of single omega-3 fatty acid is 70% or more and a content of single omega-3 is 80% or more. High-content and high-purity omega-3 triglyceride according to the present invention may be advantageously used as a medicine, food, a main raw material or supplementary material of food, a food additive, functional food or beverage.
