METHOD FOR PROMOTING DE NOVO ROOT REGENERATION OF ULTRADIAN RHYTHM-BASED PLANTS

Abstract:
Ultradian rhythms (URs) have a period of less than 24 hours. However, the regulation and function of these URs are not clear. The inventors of the present invention have confirmed circadian-independent URs having a period of less than 3 hours in excised Arabidopsis leaves. Gene expression and functional analyses suggest that URs are a positive regulator of de novo root regeneration (DNRR). 4,073 genes, including phytohormone- and DNRR-related genes representing URs have been identified by transcriptomic analysis. The inventors of the present invention have identified regulator of ultradian rhythm (rou) mutants using forward genetic screening and have confirmed that enhancer of ABA co-receptor 1 (EAR1) is essential in UR cycling. rou1 mutants exhibited a lack of URs, and reduced root regeneration and related gene expression. Hormonal treatment changed EAR1 levels in post-transcriptional and post-translational processes, which means that EAR1 mediates dynamic hormone-related gene expression in excised leaves, thus producing URs and promoting DNRR. According to the present invention, it can be confirmed that leaf excision induces URs, leading to DNRR through hormonal regulation, which is a possible survival mechanism in plants.
