Abstract:
The purpose of the present invention is to provide a simple means whereby the absolute quantities of two or more kinds of target nucleic acids in a sample can be simultaneously determined. Provided is a method for determining the absolute quantities of two or more kinds of target nucleic acids in a sample, said method comprising: (a) a step for mixing the sample with a known quantity of a control nucleic acid; (b) a step for co-amplifying all of the target nucleic acids in the sample, specific nucleic acids that are collectively amplified with the target nucleic acids and the control nucleic acid; (c) a step for determining the total quantity of all of the target nucleic acids in the sample and the specific nucleic acids using, each as an indication, the total quantity of all of the target nucleic acids and the specific nucleic acids having been amplified and the quantity of the control nucleic acid having been amplified; and (d) a step for calculating the absolute quantities of the individual target nucleic acids in the sample depending on the occupancies of the individual target nucleic acids relative to the sum of all of the target nucleic acids and the specific nucleic acids, said occupancies being calculated using, each as an indication, the total quantity of all of the target nucleic acids and the specific nucleic acids having been amplified and the quantity of the target nucleic acids having been amplified.
