Abstract:
The present invention belongs to the field of genetic engineering and provides a modified CaPif1 helicase, a complex comprising the helicase, a construct comprising the helicase, a method for using the helicase or the complex or the construct to represent a target polynucleotide, and use. The provided modified CaPif1 helicase and the complex and the construct thereof all can control the movement of a polynucleotide when passing through a biological nanopore. Especially when the length of a polynucleotide chain is increased, the modified CaPif1 helicase and the complex and the construct thereof can still stably control the polynucleotide to move without falling off from the polynucleotide. Further provided is a specific CaPif1 helicase mutant, which improves the capability thereof in controlling the displacement of the polynucleotide when passing through the nanopore.
