Abstract:
The present invention relates to a molecular peptide mutant, and an amino acid sequence as shown in SEQ ID NO: 1. By means of the invention, SpyCatcher is designed and modified on the basis of not affecting the formation of isopeptide bonds, to obtain the molecular peptide SpyCatcher-21 which has a pH stimulus response, Pro is introduced into the key loop of SpyCatcher-21 by analyzing a crystal structure to reduce the flexibility of the loop, so as to obtain the mutant SpyCatcher-21_A82P, and the linking efficiency is increased, and same can be used to achieve double-enzyme catalysis by changing the pH of the environment to obtain different degrees of coupling according to objective needs; or a three-enzyme-coupled catalytic system is obtained by means of electrostatic interaction with a positively charged enzyme.
