Abstract:
A multi-omics library construction method, a detection method, and a related kit. The multi-omics library construction method comprises: placing a slice sample on a chip having a PolyA capture function for protein capture and mRNA capture, and carrying out protein capture by using a nucleic acid-coupled antibody; respectively carrying out reverse transcription and complementary strand synthesis on the captured mRNA and a nucleic acid sequence on the nucleic acid-coupled antibody to respectively obtain cDNA and a complementary strand of the nucleic acid sequence; and carrying out library construction on the cDNA and the complementary strand of the nucleic acid sequence to obtain a transcriptome library and a proteome library of the slice sample. An antibody coupled nucleic acid having PolyA is used, the PolyA of the antibody coupled nucleic acid and mRNA binding to an antigen protein are captured at the same time by using a PolyT structure on a chip, simultaneous construction of a proteome library and a transcriptome library and subsequent co-location detection are implemented, and therefore, full-tissue view field detection of a transcriptome and a proteome are achieved.
