Abstract:
A mutant glycine oxidase has an amino acid sequence produced by substituting at least one amino acid residue in the wild-type amino acid sequence derived from a thermophilic bacterium belonging to the family Bacillus by another amino acid residue, and has the following enzymatic properties: molecular weight: 40,000 ± 2,000 daltons as measured by SDS-PAGE; optimum temperature: 45°C as measured in the presence of a pyrophosphoric acid salt under the condition of pH 8.5; optimum pH: pH 8.0 as measured in the presence of a pyrophosphoric acid salt under the condition of 37°C; thermal stability: stable at a temperature up to 70°C as measured in the presence of a pyrophosphoric acid salt under the condition of pH 8.5 while retaining for 1 hour; pH stability: stable at a pH value falling within he range from 5.5 to 10.0 as measured in the presence of a pyrophosphoric acid salt under the condition of 4°C while retaining for 24 hours; specific activity: 1.2 unit/mg or more; and a kinetic constant Km: 0.2 mM or less.
