Abstract:
Disclosed are a mutant transposon vector and a method for introducing a foreign gene into a cell. A mutant transposon vector having a nucleic acid fragment inserted therein, the nucleic acid fragment having the properties (i) and (ii) or the properties (i) to (iii) below; a foreign gene-expressing cell which is prepared by introducing the mutant transposon vector into the chromosomal DNA of an established cell or a cell capable of being individuated and then additionally introducing a foreign gene-expressing cassette into a recombinant system (e.g., Cre-Lox system); a recombinant animal created by using the cell (in the case where the cell capable of being individuated is used); and a method for producing a foreign protein from the cell or the recombinant animal: (i) comprising a 5'-TIR sequence and a 3'-TIR sequence of transposon gene; (ii) having a sequence at which a recombination reaction occurs inserted between at least one set of DR regions of two sets of DR regions present in a 5'-TIR sequence or a 3'-TIR sequence of transposon gene; and (iii) having a restriction recognition site or a foreign gene-expressing cassette inserted between a 5'-TIR sequence and a 3'-TIR sequence of transposon gene.
