Abstract:
The invention concerns a gene coding for a novel human GnRH receptor (gonadotropin-releasing hormone receptor), hereafter referred to as GnR-R2. The DNA sequences of the invention can be placed in expression systems so as to produce GnRH-R2 and/or cell lines expressing GnRH and preferably responding to GnRH-induced transduction signals. Such cell lines can advantageously be used for screening and identifying novel GnRH agonists and antagonists. In other aspects of the invention, the DNA encoding GnRH-R2, antisense oligonucleotide sequences, GnRH-R2 expression products, antibodies against such products can be used in diagnostic methods or for treatment of disorders related to an abnormal expression of GnRH-R2, for example an overexpression, a subexpression or an expression of a mutant receptor. Transgenic animals containing the GnRH-R2 transgene can be used as models for evaluating analogues of GnRH in vivo.
