Abstract:
Disclosed is a phase modulation fluorescence differential microscopic imaging apparatus based on an electro-optical modulation technology. The phase modulation fluorescence differential microscopic imaging apparatus comprises an illumination system for generating an excitation light beam, a detection system for collecting a fluorescence signal emitted by a sample, and a computer for control and signal processing. The illumination system comprises, arranged in sequence, a laser device, used for emitting a laser beam; an electro-optical modulation module, used for controlling rapid conversion of the laser beam between right-handed circularly polarized light and left-handed circularly polarized light; and a phase modulation device, used for performing phase modulation on the circularly polarized light to form a solid light spot and a hollow light spot which illuminate the sample and excite fluorescence. Also disclosed is a phase modulation fluorescence differential microscopic imaging method based on the electro-optical modulation technology, rapid conversion of solid and hollow light spots is achieved, and an imaging speed of a fluorescence differential microscopic imaging system is increased to a great extent.
