Abstract:
Polyethylene glycol derivative modified interleukin-12 (IL-12), a preparation method therefor and an application thereof. The polyethylene glycol derivative modified IL-12 is obtained by performing a modification reaction on a polyethylene glycol derivative and IL-12 Active groups of the polyethylene glycol derivative comprise one or more among a aldehyde group, ketone group, maleimide group, vinyl sulfone group, terminal alkyne group, succinimide group, p-nitrophenyl group, isobutyl chlorocarbonate group, halo group, thio group and carboxyl group. Active groups of the IL-12 comprise one or more among an N-terminus α-amino group, ε-amino group of a lysine side chain and free sulfhydryl group of cysteine. IL-12 that has passed through polyethylene glycol derivative modification may balance the drug activity and drug toxicity of IL-12, and a larger dose may be used in a single dose. In addition, the IL-12 may maintain an effective blood concentration in the body for a longer period of time without causing serious toxic side effects, which is beneficial for the continuity of treatment and improves the treatment effects of diseases.
