Abstract:
A process for preparing optically active clopidogrel and its analogues methyl tetrahydrothienopyridine acetate of formula (XII), uses halophenylacetonitrile (VIII) as starting material, tetrahydrothienopyridine acetonitrile (IV) and tetrahydrothienopyridine acetate (V) as key intermediates to obtain clopidogrel (X=2-Cl) and its analogues tetrahydrothienopyridine acetate racemic compounds (I); and resolves the said racemic compounds (I) to obtain optically active clopidogrel and its analogues tetrahydrothienopyridine acetate compounds of formula (XII). Clopidogrel prepared by the present invention is a novel, highly efficient and safe agent against platelet aggregation. Taking advantage of system technologies such as racemization of undesired optically active enantiomer, recovery, recycle and comprehensive utilization of resolving agent and so on, the present invention provides an economic benefit and is suitable for industrial scale production. wherein: X represents hydrogen, fluorine, chlorine, bromine or iodine atom; Y is Br, Cl or ester group; M is an alkali metal ion.
