Abstract:
The present invention sets forth a preparation method for a drug-linker MC-MMAF used as an antibody-drug conjugate, and an intermediate thereof. The preparation method of the present invention improves the reactivity of an N-terminal, thus effectively controlling the occurrence of a racemization reaction; and does not directly utilize the toxin MMAF, but rather uses a lower-toxicity fragment peptide, thus reducing the operational difficulty of mass production; not requiring reverse phase preparation, and being operationally simple.
