Disclosed are a preparation method of soil reinforced with a bio-binder using a microorganism, and soil prepared thereby, wherein the soil is harmless to the human body and does not cause environmental pollution, and the strength and wind resistance of the soil are sufficient. The method and soil is prepared by combining sand comprising silica and alumina as main components, a microorganism producing urea, urea, a calcium ion, and a polymer fiber, wherein the amount of the polymer fiber is 0.05-5 wt% on the basis of the amount of the sand. In addition, the microorganism preferably provides a CaCO3 production rate of 1-7×10-9 (g CaCO3 ppt cell-1 hr-1) in the standard state.
