Abstract:
Preparation and use of a thermophoric β-glucosidase, belonging to the technical field of genetic engineering and fermentation engineering. By constructing a recombinant fungus, β-glucosidase TpBgl3A derived from thermophilic fungus Talaromyces piceae is heterologously expressed, and the recombinant fungus can produce the enzyme up to 2324 U/mL in a 3.6L fermenter. The beta-glucosidase TpBgl3A produced can prepare an gentiooligsaccharide with a high conversion rate using glucose as a substrate at a relatively low enzyme addition amount, thereby significantly reducing production costs; and in a reaction system using glucose and cellobiose as substrates, has a conversion rate of the gentiooligsaccharide up to 26.2%, having the potential of industrial production of the gentiooligsaccharide.
